Tom Cech will be hosting this visit. \nAbstract: \nTelomeres protect chromosome ends from inadvertently activating DNA damage signaling and repair and they provide mechanisms for their maintenance by semiconservative DNA replication\, telomerase and homologous recombination. DNA damage signaling and telomere maintenance pathways must be fine-tuned at chromosome ends to be only active when appropriate. This requires the interplay of a large set of proteins that associate stably or transiently with chromosome ends. \nTo obtain insights into the molecular networks\, our laboratory established methods to decipher telomeric chromatin composition in human cells under normal and mutant conditions and during DNA replication. First\, I will report on telomeric chromatin changes that occur upon loss of the single strand telomeric DNA binding protein POT1. We find that human POT1 prevents the pathological engagement and hyperactivation of the homologous recombination machinery at telomeres. The suppression of homology directed repair is further supported by another OB-fold containing protein\, which we identified at telomeres. Second\, I will present on studies of the telomeric long noncoding RNA TERRA\, which in contrast to POT1\, stimulates homologous recombination. Telomere association of TERRA requires its repetitive UUAGGG-telomeric repeats and it relies on formation of DNA/RNA hybrid structures. We identified machineries that regulate TERRA recruitment. Curiously\, TERRA localization to telomeres is promoted by homologous recombination proteins.
